USE OF HIGH-PRESSURE LIQUID CHROMATOGRAPHY FOR QUALITY SURVEILLANCE OF A SUBSTANCE AND ALSO SYNTHESIS STAGES AND DYNAMICS OF AN ANTIVIRAL DRUG GAMMAVIR article

The article presents the results of high-pressure liquid chromatography usage for quality surveillance of a substance and stages of antiviral drug Gammavir synthesis dynamics, which represents threecyclic structure and has set of isomers. Were investigated: synthesis dynamics of intermediate and finished products and presence of admixings in finished products. The investigation has shown, that the synthesis of intermediate compound - diglucopyranosyl-ortho-diazine corresponds with forecasted synthesis scheme: reaction mixture contains three separate substances-isomers. These substances are noncondensated intermediates of the reaction. Gammavir substance corresponds to conditions of pharmacopoeia article, though contains admixings but within acceptable limits. The last stage of Gammavir synthesis research has shown, that the mechanism of synthesis is much more complex than that it was first suggested: 10 minutes after the reaction start the reaction mixture contains more than 18 intermediate compounds, which are vanishing during Gammavir synthesis. Thus, there could be made a conclusion about expediency of liquid chromatography usage during development of industrial synthesis technologies for medicinal substances, intermediate synthesis stages control, synthesized substances purity and quality analysis, especially for those substances, which contain several isomers and synthesis intermediates.
